    Mutations in RAC2 have been identified in a small number of patients giving rise to different forms of primary immunodeficiencies ranging from granulocyte defects caused by dominant negative mutations to combined immunodeficiency due to dominant activating mutations.
    It appears therefore that activating Rac2 mutations have a prevalent effect on lymphocytes, giving rise to a CID like phenotype, whereas the effect of the dominant negative mutations in Rac2 is mainly restricted to the granulocyte subsets.
    The main differences between Rac1 and Rac2 are near the C-terminal prenylation consensus site (CAAX box). This region is composed of six basic amino acids (KKRKRK, designated as a polybasic motif) in Rac1. In Rac2, three of these six amino acids are replaced by neutral amino acids (RQQKRP).
    Here we have presented evidence that the specificity of function of Rac2 versus Rac1 in primary neutrophils is regulated at least in part by distinct subcellular distribution of these GTPases that is in turn determined by the C-terminal RQQKRP sequence and the aspartic acid at position 150 of Rac2.
